Please note that because of the design and function of a molecular beacon probe, it may be possible to see different states of fluorescence under different conditions. Lower temperatures tend to put the beacon probe in what is known as a "closed state" which gives off little to no fluorescence.

In order to clearly see any fluorescent label, a certain temperature may be required. It is possible to control the temperature threshold required to see this fluorescence by lowering or raising the GC content, and the length of the stem sequence. In most cases, a Midland Certified Reagent Company technician will make this decision for you, unless otherwise specified.
